    Klingt sehr interessant, vielleicht kann es ja mal jemand mit mehr Ahnung kommentieren:
    Researchers at Western University in London, Canada, have identified a new genetic mutation for amyotrophic lateral sclerosis (ALS), opening the door to future targeted therapies. Dr. Michael Strong, a scientist with Western's Robarts Research Institute and Distinguished University Professor in Clinical Neurological Sciences at the Schulich School of Medicine & Dentistry, and colleagues found that mutations within the ARHGEF28 gene are present in ALS. When they looked across both familial and sporadic forms of the disease, they found that virtually all cases of ALS demonstrated abnormal inclusions of the protein that arises from this gene.

    Seminal findings
    Josef Penninger, scientific director at the IMBA and last-author of the study, is excited about the researchers' findings: "This surprising discovery of a role of CLP1 in the onset of motor neuron diseases is an entirely new principle in how RNA talks to oxidative stress. Nearly all genetic mutations found in ALS patients affect either RNA metabolism or oxidative stress, suggesting a possibly unifying principle for these diseases. Our work may have revealed the 'missing link' in how these two biological systems communicate and trigger incurable diseases like ALS."
    Stefan Weitzer sees tremendous potential for these findings: "We've discovered a new mechanism that leads to the death of motor neurons. If this holds true for other neuronal disease, our results could be one day used to drive the development of treatments for previously incurable diseases. In our work we also describe how the p53 protein regulates the loss of motor neurons. Removing p53 saves mice with CLP1 mutations from certain death." If scientists are successful in applying these findings to people, the researchers may have discovered a treatment approach to cure ALS and similar diseases. The authors, however, caution that more studies will be needed to translate their findings to human medicine. +++

        es ist gut wenn jemand Informationen sammelt. Damit ist uns allen sehr geholfen, da man immer wieder Hoffnung schöpft.

          Proteinfaltung - Gen identifiziert

          Packard Researchers Identify the Function of an ALS-linked Gene

          Schon länger wird im Zusammenhang mit ALS eine gestörte Proteinfaltung oder Mitochondrienfunktion diskutiert.
          Forschern ist jetzt gelungen, die normalen Funktionen des Gens VCP zu identifizieren. VCP hat eine Vielzahl von zellulären Funktionen, unter anderem auch für den Erhalt bzw. die Reparatur von Mitochondrien (Kraftwerke der Zellen). Eine Mutation dieses Gens wird mit verschiedenen Multisystemerkrankungen, wie z. B. Morubs Paquet, Einschlusskörpermyositis oder auch frontotemporaler Demenz in Verbindung gebracht, allgemein auch als Proteinopathien bezeichnet.
          Im Experiment wurde dieses Gen bei Fruchtfliegen verändert, die in der Folge Symptome entwickelten, wie sie bei ALS üblich sind.
